  Serial Analysis of Gene Expression (SAGE) is a powerful strategy to study gene expression profiles, in specific tissues, at certain developmental stages, or in the process of cell responses to environmental stimuli. SAGE is of advantages to detect the transcripts and profiles of low-expressed genes.
  B. napus immature seeds at 5 and 9 days after pollination (DAP) were collected and used to construct SAGE library. Totally 68,716 tags were obtained after sequencing 1344 clones, of which 23,895 are unique (34.78% of total sequenced tags, Figure 1, Table 1) and 15123 tags (63.28% of total unique tags) are detected with single copy, and rest of them (8772 tags) are with more than one copy (the maximal copy number is 645, Table 1). Based on obtained total and unique tags, a double-reciprocal plot and linetype-regress analysis reveal the presence of ~35,000 expressed transcripts during B. napus early seed development.   Based on the virtual tags derived from GeneBank and full-length cDNA from our cDNA library, we constructed the "tag-to-gene mapping database" for B. napus and Brassica including B. oleracea and B. rapa. Analysis against this database resulted in the match of only 2.1% (502 tags to B. napus) and 3.6% (860 tags to Brassica) of total unique tags, respectively.
